Comparison of urinary proteome in the first two days after mating in male rats
摘要: 目的:比较雄性大鼠交配行为后前两天尿液蛋白质组变化。方法:收集Sprague-Dawley大鼠交配当天的尿液样品和交配隔天的尿液样品,通过高效液相色谱串联质谱联用(LC-MS/MS)的非标记定量蛋白质组学技术进行分析,筛选尿液蛋白质组的差异蛋白(FC>1.5 或<0.67,P<0.050)进行蛋白质功能和生物学通路分析。结果:大鼠交配隔天与交配当天尿液蛋白质组比较可以鉴定到43个差异蛋白。通过Uniprot数据库和Pubmed数据库检索差异蛋白和相关文献报道,近三分之二的差异蛋白与精子发生相关。结论:大鼠交配隔天相较于大鼠交配当天,尿液蛋白质组就发生了变化,部分变化蛋白的已知功能与精子发生相关。
Abstract: Objective: To compare urine proteome changes in the first two days after mating behavior in male rats.Methods: The urine samples of Sprague-Dawley rats on the day of mating and the day after mating were collected and analyzed by non-label quantitative proteomics by high performance liquid chromatography-tandem mass spectrometry (LC-MS/MS), and the differential proteins (FC > 1.5 or Results: 43 different proteins were identified in the urine proteome between the day after mating and the day after mating. By searching the Uniprot database and Pubmed database and related literature reports, nearly two-thirds of the differential proteins were associated with spermatogenesis.Conclusions: The urine proteome changed the day after mating compared to the day after mating, and some of the known functions of the changed proteins were associated with spermatogenesis.
